Perfusion is a continuous process in which a suspension cell culture is continuously supplied with fresh medium to the bioreactor while spent culture media is continuously harvested. The cells are continuously filtered from the harvest stream and returned to the bioreactor to maintain a constant culture volume. This continuous process allows the cells to reach high densities as densities of 15-20 millions cells/mL can routinely be reached and maintained for at least 2 weeks. This results in very highly productive cell culture process that can produce for a longer period of time as opposed to batch or fed-batch cultures.
Perfusion has become regarded as an optimal process option when the stability of the protein is low or if the expression of the cell line is minimal so that a constant output becomes more economical.
Novasep has mastered the development of novel perfusion cell culture processes using a variety of separations technologies including sonoperfusion, Alternative Tangential Flow perfusion (ATF) and hollow fiber perfusion. We also have bioreactors that are equipped with the latter two devices to be used in adherent cell culture performed on microcarriers.
During sonoperfusion the cells are retained inside the bioreactor by using a BioSepTM device. The BioSep TM is an acoustic device that utilizes megahertz range ultrasonic waves to separate suspended cells from cell culture medium allowing the target protein to be continuously secreted. We perform sonoperfusion mainly at the 10 L scale.
The ATF™ System is based upon the technology of an alternating tangential flow over a hollow fibre, created by the action of a moving diaphragm., We have a successful track record with ATF perfusion from 10 L to 80 L scale.
